HAT系统为次黄嘌呤(H)、氨基喋呤(A)和胸腺嘧啶核苷(T)的缩写。它是根据嘌呤和嘧啶生物合成途径设计的分离杂种细胞特殊的培养基。细胞内核苷酸的生物合成有两条途径:一条是主要途径,该途径中叶酸及其衍生物是必不可少的,氨基喋呤可抑制二氢叶酸还原酶活性,从而阻断细胞中DNA的合成;另外一条是补救途径,该途径需要两种酶参与。

一种是HGPRT酶(次黄嘌呤磷酸核糖转移酶),另一种酶是TK酶(胸腺嘧啶核苷激酶)。它们可以分别利用次黄嘌呤催化产生的肌苷酸及胸腺嘧啶核苷催化产生的脱氧胸苷来合成DNA。H、T为应急途径时提供外源核苷酸“前体”。在氨基喋呤存在时,阻断了核苷酸的从头合成IMP的途径,细胞只能通过HGPRT使次黄嘌呤酸化形成次黄苷酸,再依次转化为AMP及GMP,即通过“补救途径”依赖外源来合成核苷酸。因此,一个HGPRT-或TK-的细胞株不可能在含HAT的培养液中生长。

但这样的细胞与能提供HGPRT或TK酶的细胞相融合,则杂交细胞能在含HAT培养液中存活下来。如果融合细胞的亲本之一是一个能在体外长期生存的骨髓瘤细胞系,而另一个亲本细胞是体外增殖能力有限的正常细胞。

如将长期培养系突变为HGPRT或TK酶缺陷型的细胞系,在和正常细胞融合后,未经融合或自身融合的骨髓瘤细胞在HAT培养液中死亡;未融合或自身融合的正常细胞在体外生存能力有限最终也死亡。因此存活下来的只有融合细胞。
